Vestibulodynia (VBD) is a condition of persistent vulvar pain localised to the vaginal vestibule; this common disorder, affecting approximately 10-18% of women at various stages of life, typically manifests as a burning, itching or uncomfortable sensation upon contact with the vestibular area (e.g. during sexual intercourse or use of tampons). VBD is characterised by hypersensitivity of vestibular nerve fibres with a neuroinflammatory origin, often associated with dysfunction of the pelvic floor muscles, primarily of the hypertonic type.
A team led by Prof. Filippo Murina at the Lower Genital Tract Pathology Service at the Vittore Buzzi Hospital in Milan (Italy) conducted a randomised double-blind placebo-controlled study on 40 women with VBD to evaluate the efficacy of ALYVAL MED over a 60-day period.
Participants' symptoms, such as pain and dyspareunia (primary efficacy endpoint), muscle hypertonus (clinical assessment) and vestibular nerve endings sensitivity (measured using the Neurometer device), were assessed at day 0 and day 60.

A 70% reduction in pain and a 20% reduction in dyspareunia was observed in the ALYVAL MED-treated group compared to the placebo-treated control group.

Current Perception Threshold (CPT) refers to the minimum intensity of an electrical stimulus required for nerve fiber perception, including vestibular nerve fibres. Stimuli are delivered at three frequencies (2000 Hz, 250 Hz, 5 Hz) with a gradual increase in intensity until the minimum perception threshold is reached.
In the ALYVAL MED-treated group, an approximately 20% increase in the pain threshold was observed compared to the placebo-treated control group; this data suggests a reduction in nerve sensitisation, one of the primary symptoms of this pathology.
These are preliminary results that indicate even greater improvement at the conclusion of the study.
